Although the components in natural propolis are very complicated, most of the effective components can be well dissolved in ethanol and sodium hydroxide solution and only a few substances can be dissolved in nonpolar solutions, such as beeswax. Most of the effective substances in propolis can be extracted by mixing petroleum ether with ethanol, but the process is complicated and only suitable for large-scale production due to process limitations. The polarity of acetone is close to that of ethanol, but the extract can not be directly made into tincture as ethanol extraction, and the extract is used in medicine, food, cosmetics, and so on. The following introduces several feasible purification technologies of bee glue.
1. Two-phase solvent extraction method of ethanol and petroleum ether Put the pulverized natural propolis powder in an extractor, add 4 times 90% ethanol and 1 time 60-90 petroleum ether (weight: volume), stir, and extract at a constant temperature of 40℃, repeat the above three times, combine the extracts, separate layers, cool, filter and concentrate under reduced pressure. Mix fat-soluble and alcohol-soluble substances to obtain pure propolis. It was confirmed by experiments that the volume-weight ratio of petroleum ether and ethanol solution to propolis was in the range of 1∶4∶1, the extraction rate was high and the components were complete at 40℃. However, because the equipment and process are more complicated than single solvent extraction, it is suitable for large-scale production.
Second, sodium hydroxide solvent extraction method Add 1% and 2% sodium hydroxide solution to the crushed natural propolis 10 times, fully stir it, filter it immediately after it is dissolved, then add 5%HCL aqueous solution to acidify the propolis, wash it with water, and then injection mold it. This method is difficult to master in technology, and there will be too long a time and too high a temperature in the process of dissolution and acidification with an alkali solution, which will make the separated propolis appear as slag. The analysis shows that, although sodium hydroxide solution has good dissolution characteristics for the effective components of propolis, it can not reduce all the components after acidification, and some irreversible reactions such as foreign body transformation and partial saponification into acidity occur. Therefore, it is inappropriate for sodium hydroxide to be used as the solvent for all effective components of natural propolis.
3. Ethanol solution impregnation method The pulverized propolis powder was added to 5 times of ethanol aqueous solution with concentrations of 75%, 85%, and 95% respectively. Soak at room temperature for 24 hours, take out the extract, add it twice, cool the combined extracts to below 5℃, filter, and recover the solvent under reduced pressure. Its concentrate is dark brown to black pure propolis.
