Preparation method
Ferulic acid can be obtained from plants in three ways: from its binding to some small molecules, from plant cell walls, and through tissue culture. Ferulic acid in plants is mainly cross-linked with polysaccharides and lignin through ester bond or self-esterification or etherification to form diferulic acid. It is generally used to break the ester bond to release ferulic acid by alkali method and enzyme method, and then extract with appropriate solvent.
1. Alkalysis
Ferulic acid from cell wall can be released by using 4% sodium hydroxide to react at room temperature for 24h under the condition of nitrogen. Recent studies have found that most ferulic acid can be extracted from wheat bran in a short time by increasing the extraction temperature and adding suitable protective agent. Oshi et al. use low concentration of sodium hydroxide solution, can release most ferulic acid in wheat bran at appropriate extraction temperature, adding sodium sulfite in the extraction process can increase the recovery rate of ferulic acid. Due to the complex composition of lye, especially containing pigment substances, the main separation method of ferulic acid in lye is activated carbon adsorption. The structural unit of ferulic acid is contained in glutamate, which exists in the form of ester and is easy to decompose. Therefore, it can be hydrolyzed glutamate with alkali first, and then prepared ferulic acid by acidizing method. The reaction of hydrolyzed glutamate to produce ferulic acid is convenient to operate, the yield is as high as 85.7%, and the by-product is cycloxybromeliol. Moreover, the source of glutamate is wide, the yield is large, and the price is moderate.
2. Ferulic esterase method
Ferulic esterase is an enzyme that can free ferulic acid from methyl ferulic acid, oligosaccharide ferulic acid and polysaccharide ferulic acid. Ferulate esterase is secreted by fungi, bacteria and yeast. Using Aspergillus Niger as a strain, Zhang Jing et al. prepared a mixture of ferulic esterase and arabinxylanase by liquid deep fermentation method. The mixture of enzyme preparation was applied to the destarching wheat bran, and the degradation rate of wheat bran reached 55.46% after three times of degradation.
3. Plant tissue culture method
Plant tissue culture is an important way to obtain ferulic acid. Some studies have shown that tissue culture of certain plants can yield higher yields of ferulic acid derivatives. For example, water soluble glucose ferulate ester and sucrose ferulate ester can be obtained by cell suspension culture of sugar beet and corn, the content of which can be as high as 20.0unol/g callus (dry weight). The content of ferulic acid in the direct extract is relatively low, which requires further purification.
